Synthetic Studies on Heterocyclic Natural Products

Abstract: This presentation highlights recent efforts focusing on the total synthesis of biologically relevant nitrogenous natural products. Each target molecule poses chemical problems that serve as ideal vehicles for the development and application of new synthetic technology. Compounds of current interest include (i) thiopeptide antibiotics (genetic modulators and inhibitors of protein synthesis); (ii) polyoxazole natural products such telomestatin (potent and selective inhibitor of human telomerase); (iii) substances incorporating medium-ring nitrogen heterocycles or surrogates thereof such as mitomycinoids (antitumor agents); and (iv) spirocyclic alkaloids (immunusuppressants; cytotoxic agents; ion-channel blockers). The methodology aspects of each project will be discussed with an emphasis on transformations that rely on the use of hypervalent iodine reagents.
