Inhibitor of zymogen synthesis: WARFARIN
Mechanism of Action
-
inhibit epoxide reductase (hydrase), which is the most important enzyme responsible for regenerating reduced vitamin K
- NOT DIRECT INHIBITION of the post-translational modification step whereby carboxylated Gla residues are generated
- ultimately decreases synthesis of the vitamin K-dependent clotting factors II, VII, IX, X
- also decreases the activity of protein C (activated by thrombin)
→ responsible for some side effects
